What semaglutide is

Semaglutide is a 31-amino-acid synthetic analogue sharing roughly 94% structural homology with native human GLP-1 (7–37). Two engineering changes make it useful as a research tool: substituting alanine at position 8 with 2-aminoisobutyric acid (Aib) blocks cleavage by dipeptidyl peptidase-4, and a C18 diacid chain attached via a spacer to lysine at position 26 enables reversible albumin binding and a substantially extended half-life.
